I work with rat cardiac membranes. I am trying to filter the membrane homogenate using standard cheesecloth. When I filter the homogenate, I lose most of my original volume. I have tried reducing the area of the cheesecloth as well as prewetting the cheesecloth with buffer before filtering but I am still losing as much as 75% of my volume.

Do you have any suggestions as to how I can resolve this problem?

I'm not sure why you're using cheesecloth or how large your sample is (probably pretty small).

If you're pre-wetting the cheesecloth, make sure it is saturated (as wet at the start of the filtration as it will be at the end). You might also check to be sure that you're homogenizing the membranes completely (anything not completely ground up should remain behind and detract from your yield).

If possible, you may want to try a hydrophobic filter. You can get Teflon and polyolefin woven filters with very small openings. These will not absorb any of your homogenate like cotton does.
