After rupturing bacterial cell, I used a 0.1 mciron size of hollow fiber to separate cell debri and cell soluble fraction that contains protein of interested. The MW of protein is 100 kDa. Unfortunately, more than 65% of desired protein is in the retentae size. My goal is more than 80% in the permeat size. Any suggestion?
