My liposomal suspension containing Timolol maleate entrapped in small unilamellar vesicles (nearly 50 nm diameter) and I want to separate untrapped timolol maleate from the trapped one to obtain the SUVs containing the trapped timolol maleate only. How can I do this?
how can i determine the drug size in daltons?
this separation will be called fiteration or dialysis?

The drug size is the same as its molecular weight.

This process will be either filtration, if it is under pressure, or dialysis, if it is passive.

Let's see: Timolol MW 316, Maleic Acid MW 116, so Timolol Maleate should be around 423.

The separation you choose will depend upon your sample size and the stability of the drug in the vessicles.

If your sample size is in the milliter range (say 1 to 200 mL) and the vessicles are stable so that the trapped drug is not in equilibrium with the free drug, then dialysis is your best bet. Use good ole Spectr/Por 4 (http://www.spectrapor.com/dialysis/spectra_por_4.html) in a size that is convenient. (Be sure to dialyze against a solution with an osmotic pressure similar to that of your sample.)

If the vessicles are not stable, or there's an equilibrium, or if your sample is much larger or smaller, then the process is more complicated.
